Water conservation measures to slow down the runoff and increase the water supply in the floodplain of Kozí stream

This thesis deals with the variety of measures in the floodplain of the piped Kozí creek, which aim to improve its morphological and ecological condition, slow down the runoff from the basin and increase the retention capacity of its surroundings. The thesis contains theoretical knowledge about revitalization, renaturation and restoration of water reserves in the landscape, which is then applied to concrete solutions. Associated with this topic is also the drainage of the landscape and an assessment of historical drainage interventions from a contemporary perspective, within which the thesis addresses theoretical and practical dealings with drainage structures that are no longer needed or no longer functional. The thesis includes the implementation of watercourse monitoring according to the HEM methodology, which serves as a tool for assessing the hydromorphological condition of watercourses. The evaluation of the monitoring is then used for the design of individual measures.
Effects of restoration and renaturation measures applied on watercourses

This work focuses on the effects of restoration on watercourses, in particular on a planned restoration of Klíšský brook in Ohře basin and a finished restoration of Rokytka in Běchovice. The main goal is to use the methods HEM and MoRPh for evaluation of hydromorphology in both areas. In case of Klíšský brook, HEM is used to estimate ïts future condition after restoration. Another goal is to assess and propose a vegetation structure for the of Klíšský brook restoration, then to describe it on Rokytka. Research on the topic of river restoration, their goals and effects is included, as is an introduction to in the Czech Republic less known method MoRPh. The results of hydromorphological and vegetation structure assessment are introduced. Both utilised methods show adverse hydromorphological conditions on Klišský brook in comparison to Rokytka. Vegetation also shows higher diversity around and in Rokytka's channel. This work is a base for further monitoring of hydromorphology and vegetation evolution in both areas. Restoration and renaturation in the Klíšský Brook catchment

This bachelor's thesis presents a revitalization and renaturation study of a small watercourse in rural area. Specifically, it is the Klíšský stream, which is a tributary of the Bílina River. Among the main goals of the bachelor thesis is the development of possible proposals for possible revitalization and renaturation measures with the aim of improving the hydromorphological quality of a small watercourse. During the development of the revitalization plans, a total of 18 cross-section profiles and one longitudinal profile were surveyed within the geodetic survey, in which specific revitalization measures were proposed. Within the evaluation of the characteristics of the study area, analyses of the granular composition of the sediment of the bottom were carried out, an evaluation of the property conditions was carried out and also flow monitoring was carried out during the periods of field surveys. In the revitalization modification proposals, the existing capacity in the targeted transverse profiles was also calculated. By synthesis of all the obtained and available information, proposals for measures to improve the overall condition of the water course were carried out, in the section the water surface in the form of a pool and wetlands in the area of the non-functional hydromelioration network...

The main objective of this thesis is to evaluate the impact of the revitalization of the Stropnice River on the amount of water retained in the landscape. The theoretical part of the thesis focuses on the description of the revitalizations and the water reten-tion in the landscape. The practical part focuses on the description of the selected area, the description of the revitalisation of the given section and the comparison of the amount of water in this section before and after the revitalisation. The revitali-sation has benefited not only the stream, the length of which has been considerably extended by the meanders in the section in question, but also the wider river area, which is currently showing signs of waterlogging.
Preparation of revitalization study for small water course

This diploma thesis deals with returning of modified troughs of small watercourses to the natural state - revitalisation. The general idea of the river beds and water environment revitalisation will be explained in the theoretical part. It will also deal with its history, development and practical application. The practical part will contain an own project creation. It is a project of the adjusted small watercourses bed revitalisation in the level of study. A section of a fine river was used for this thesis. It is called Žďárský Stream and it belongs to the cadastral area Žďár near the Kaplice. Individual parts of the river and its basin will be mapped in this work. It will also consider the hydrological and geographical conditions and it will propose a technical solving for a local revitalisation of the adjusted river bed. It will ínclude natural factors support, the ecological-morphological improvement, the migrational permeability and other links to the river environment.
Preparation of water course revitalization study

This diploma thesis deals with the design of ox-bow lake revitalization measures, located southwest to the city Strakonice. Due to its charakteristics, ox-bow lake represents high-value biodiversity area. Recently, due to a disturbation in the aquatic regime, this area is threatened in sediment issue. Practical part of the diploma thesis is focused on the sediment and water level research and monitoring of plant and animal species. Obtained data result in particular revitalization measures.
NMR study of the extracellular part of the mouse Nkr-p1b receptor from natural killer cells

Protein Nkr-p1b is a surface receptor of cytotoxic NK cells, that mediates inhibitory signal toward the body's own cells. In this study, the ligand binding domain of the mouse protein receptor Nkr-p1b (mNkr-p1b LBD) was prepared by recombinant expression in E. coli cells. Isolated protein was subsequently used for NMR structural analysis. Prediction of protein secondary structures ratio was carried out using three different methods (CD, PSIPRED and TALOS). Results correlate well with the structure of CTLD domain, that plays a key role in ligand binding and thus to function of Nkr-p1b receptor. We managed to prepare this protein in a form suitable for NMR experiments. Based on the data obtained by NMR spectra analysis, a preliminary model of the mNkr-p1b LBD protein structure was created. However, for more precise learning of the 3D structure accurate positions of individual atoms need to be determined by other NMR spectra evaluation in the next phase. Explaining the structure of the ligand binding domain of mNkr-p1b protein could help to better understand the complex mechanism of activation of NK cell cytotoxic activity, thereby contributing to its controlled use as a therapeutic against some viral and tumor diseases.
